Tesamorelin is a synthetic analogue of growth hormone-releasing hormone (GHRH), consisting of 44 amino acids with a modification that increases its stability and half-life. It stimulates the pituitary gland to release endogenous growth hormone (GH), which in turn elevates levels of insulin-like growth factor 1 (IGF-1).
Tesamorelin is FDA-approved under the brand name Egrifta for the reduction of visceral adipose tissue (VAT) in HIV-infected patients with lipodystrophy. In pivotal Phase III clinical trials published in The New England Journal of Medicine (2010), Tesamorelin significantly reduced visceral adipose tissue by an average of 15–18% over 26 weeks compared to placebo, without adversely affecting subcutaneous fat. Improvements in triglyceride levels and markers of inflammation were also observed. Recent research has also explored Tesamorelin’s potential benefits in non-alcoholic fatty liver disease (NAFLD) and cognitive function in older adults with HIV.
